Martin & Jones is investigating the potential connection between the use of the injectable contraceptive, Depo-Provera, and the development of meningioma tumors. Reports have been linked to the long-term use of Depo-Provera and meningiomas.

While the Food and Drug Administration (FDA) has not issued a public statement or warning, in October 2024, Pfizer, the drug’s manufacturer, wrote to the national health regulatory agency in Ireland discussing the “increased risk of developing meningioma.”  Earlier this year, in March 2024, the British Medical Journal (BMJ) published a report warning that long-term use of Depo-Provera could increase the risk of developing a meningioma.

Following the BMJ publication of this report, Pfizer issued a statement acknowledging said risks and stated it was “in the process of updating product labels and patient information leaflets with appropriate warnings.”  Pfizer updated warning information for patients in Europe; however, as of December 3, 2024, Pfizer has not made label changes informing patients of the potential meningioma risk in the United States.

The Mayo Clinic explains, “A meningioma is a tumor that grows from the membranes that surround the brain and spinal cord, called the meninges. A meningioma is not a brain tumor, but it may press on the nearby brain, nerves and vessels.” 

Depo-Provera was approved by the FDA in October 1992 as an injectable contraceptive to prevent pregnancy.  There is a black box that was issued in November 2004 warning not to use Depo-Provera for more than two years for concerns over bone density issues. There is no warning, however, for the risk of developing meningiomas in the United States.  While the U.S. label has not been updated, Canadian and European labels were updated recently to inform users about the potential risk of meningiomas.

Meningiomas grow slowly and can grow for years without creating symptoms.  However, symptoms may include changes in vision such as seeing double or blurring, headaches that are worse in the morning, hearing loss or ringing in the ears, memory loss, loss of smell, seizure, weakness in arms or legs, or trouble speaking.  Doctors suggest seeking medical care for sudden onset of seizures or sudden changes in vision or memory.
